Determination of blood glucose
Principle:
Glucose in serum amples is determined according to the following reaction:
Normal value: 65-110 mg/dl (fasting blood glucose).
Reagents:
< >Standard glucose 100 mg% Working solution:Phosphate buffer, pH 6.5 225 mmol/l
Phenol 0.3 mmol/l
Amino 4-antipyrine 8.5 mmol/l
EDTA 5.0 mmol/l
Peroxidase 300 U/l
Glucose oxidase 1000 U/l
Sample preparation:
-Serum or Plasma collected in sodium fluoride, EDTA or heparin.
Sodium fluoride is added to inhibit glycolysis in RBCs by inhibiting the enzyme enolase responsible for phosphoenol pyruvate production.
Wave length: 505 nm. Measurement: against blank.

Mix, let to stand for 10 min. at 37 °C or for 20 min. at 25 °C. Measure the absorbance of Sample and Standard against blank.
Calculation:
Glucose concentation in mg/dl = A sample / A standard X 100
